I have a list of e-mail addresses in a database, and I would like to only allow users with those e-mail addresses to create an account. Is this possible? If it is possible, how difficult will this be to implement?

Along with this, you can pre-register users, and lock out anyone you have not registered. It's not exactly what you said above, but it's a side option as well.

Would this mean that we automatically create a username and password for each user?
Also, would they have to activate their accounts, or would we be able to skip that step?

You could create each user, set them to Registered, and send them a link (using a built-in feature) for them to set a new password.

OK, through Admin CP > Users > Add User
As far as I can tell, you can set up everything about that user ahead of time. Username, password, email, etc. Anything that can be edited by a user later can be used to pre-register a user, as far as I see. It shows the same form for if you were to edit a user, only the fields are blank; so for this, you could pre-register an admin or mod even.
Since you can pre-reg them with a usergroup, I think if you set them as a "user" or above group, they wouldn't have to activate. If you set them to the "Users Awaiting Email Confirmation", then they would have to.
From here, you can set up whoever you want for members, then they can come in with the account info and edit anything they see fit (such as password or email, etc).
If you know some php (seeing you already have a database, you probably already do), you can code a plugin to check if the user's email address is in the list when they're registering; and reject it if they're not.
